2012/03/06: Version 2.0, the bug with arguments calling was fixed (thank to Robert W. Baer and Johannes Schindelin).

Requires:

ImageJ 1.31i or later

Limitations:

Limited to simultaneous analysis of 2 organelles

Bugs:

Source:

IntraCell_v2.java

Installation:

Download IntraCell_v2.java to the plugins folder, or subfolder, and compile and run using Plugins/Compile and Run. Restarting ImageJ will add a "IntraCell_v1" command to the Plugins menu or a submenu of the Plugins menu.

Description:

IntraCell plugin for ImageJ program allows quantitative assessment of efficiency of intracellular drug delivery of nanoparticle-based drug delivery systems. The applied analysis algorithm is based on pixel selection and pixel-to-pixel analysis of the experimental images. IntraCell uses custom-selected thresholds to identify the borders of the studied organelles and to exclude background pixels from the analysis. The analysis requires set of images taken from the same field of view with individual images representing fluorescence of the studied nanoparticle formulation and of the individual organelles. In addition, cell borders should be identified using membrane-specific dyes, based on the cells' autofluorescence, or from differential interference contrast (DIC) image. IntraCell plugin, its validation, and application to study intracellular targeting of a novel formulation to the endoplasmic reticulum has been described in: Sneh-Edri H. and Stepensky D. 'IntraCell' plugin for assessment of intracellular localization of nano-delivery systems and their targeting to the individual organelles. Biochem Biophys Res Commun 2011, 405:228-33.
SampleImages.zip file contains the images in the required format and could be used for testing and learning the IntraCell plugin.
